In recent decades, there has been a huge level of interest in bioactive compounds fromnatural sources. The large range of plant biodiversity means that there is a huge varietyof bioactive compounds. Bioactive compounds are mostly specific secondary metaboliteswith antioxidant, inflammatory, immunomodulative potential, antimicrobial properties, etc.The bioactive compounds of classes of terpenes, flavonoids, alkaloids, coumarins, stilbenes,etc., alongside the description of some of their mechanisms of action, are important tostudy. It is important to note that the use of plant extracts complicates the identificationof the effects of antiviral, antimicrobial or other capacities of separate biologically activecompounds; additionally, the effects of activities of solute compounds in extracts can causean additive (the synergistic effect) or antogonistic effect to come into play. In that context,the current issue is open for scientific research on the description of novel isolated bioactivecompounds, as well as some of their unknown effects, which are a priority for the appliedneeds of humans in different areas of life.

Sustainable rural development in Germany was examined by linking conceptual and applied
aspects of the land and housing question, broadly considering the ownership, use, and regulation
of land. In the state of Bavaria, a new interagency initiative aims to curb land consumption by
persuading villagers to embrace rural infill development. The study explored the background debate
leading up to the Space-saving Offensive (Flächensparoffensive), the resource providers involved,
and the options for funding actual rural infill building and renovation projects. Here, space-saving
managers and other resource providers actively promote the positive societal meaning of central infill
sites in contrast to unsustainable land consumption. In addition to the communications campaign,
planning, regulatory, and funding interventions round out the multi-level initiative, as described in
this study. A modern barn reuse exemplifies the Bavarian bundle of resources, while demonstrating
how modern village infill redevelopment also contests oversimplified notions of stagnant rural
peripheries. The initiative’s focus on linking key resources and bolstering communications can be
read as validation for a more social perspective on land consumption and village infill development.

The dual-purpose German Black Pied Cattle (DSN) has become an endangered breed of
approximately 2,550 registered cows in Germany. The breed is genetically related to
Holstein-Friesian cattle because the old DSN breed contributed to the selection of
the modern Holstein dairy cow. In dairy farms, breeders aim to improve animal health
and well-being by reducing the number of mastitis cases, which would also reduce milk
losses and treatment costs. On the genomic level, no markers associated with clinical
mastitis have been reported in DSN. Therefore, we performed a genome-wide association
study on 1,062 DSN cows using a univariate linear mixed model that included a relatedness
matrix to correct for population stratification. Although the statistical power was
limited by the small population size, 3 markers were significantly associated, and
2 additional markers showed a suggestive association with clinical mastitis. Those
markers accounted for 1 to 3% of the variance of clinical mastitis in the examined
DSN population. One marker was found in the intragenic region of NEURL1 on BTA26, and the other 4 markers in intergenic regions on BTA3, BTA6, and BTA9.
Further analyses identified 23 positional candidate genes. Among them is BMPR1B, which has been previously associated with clinical mastitis in other dairy cattle
breeds. The markers presented here can be used for selection for mastitis-resistant
animals in the endangered DSN population, and can broadly contribute to a better understanding
of mastitis determinants in dairy cattle breeds.

issue in medical education. In this review, we report about the history
of the discovery of the blood groups by Landsteiner, the link of
different blood groups to certain diseases and our experiences regarding

In this article, we review the literature regarding the yield gap between organic and conventional agriculture and then reflect on the corresponding debate on whether or not organic farming can feed the world. We analyze the current framework and highlight the need to reframe the yield gap debate away from “Can organic feed the world?” towards the more pragmatic question, “How can organic agriculture contribute to feeding the world?”. Furthermore, we challenge the benchmarks that are used in present yield comparison studies, as they are based on fundamentally distinct paradigms of the respective farming methods, and then come up with a novel model to better understand the nature of yield gaps and the benchmarks that they are premised on. We thus conclude that, by establishing appropriate benchmarks, re-prioritizing research needs, and focusing on transforming natural resources rather than inputs, organic systems can raise their yields and play an ever-greater role in global sustainable agriculture and food production in the future.

The type I cGMP-dependent protein kinases (PKG I) serve essential
physiological functions, including smooth muscle relaxation, cardiac
remodeling, and platelet aggregation. These enzymes form homodimers
through their N-terminal dimerization domains, a feature implicated in
regulating their cooperative activation. Previous investigations into
the activation mechanisms of PKG I isoforms have been largely influenced
by structures of the cAMP-dependent protein kinase (PKA). Here, we
examined PKG Iα activation by cGMP and cAMP by engineering a monomeric
form that lacks N-terminal residues 1-53 (Δ53). We found that the
construct exists as a monomer as assessed by whole-protein MS,
size-exclusion chromatography, and small-angle X-ray scattering (SAXS).
Reconstruction of the SAXS 3D envelope indicates that Δ53 has a similar
shape to the heterodimeric RIα-C complex of PKA. Moreover, we found that
the Δ53 construct is autoinhibited in its cGMP-free state and can bind
to and be activated by cGMP in a manner similar to full-length PKG Iα as
assessed by surface plasmon resonance (SPR) spectroscopy. However, we
found that the Δ53 variant does not exhibit cooperative activation, and
its cyclic nucleotide selectivity is diminished. These findings support a
model in which, despite structural similarities, PKG Iα activation is
distinct from that of PKA, and its cooperativity is driven by in trans interactions between protomers.

Weed competition can severely reduce soybean (Glycine max (L.) Merr.) yields, particularly in organic systems. An efficient screening and breeding approach is needed to increase breeding progress for weed tolerance. This study sought to (i) establish a system for direct selection of competitive genotypes, (ii) evaluate genotypic differences in weed tolerance among six early‐maturing genotypes and (iii) assess the contribution of selected morphological traits to weed tolerance. A direct selection system providing two different levels of weed competition through all development stages of a soybean crop was developed, using mixtures of selected crop species as sown competitors. Two resulting mixtures induced intermediate (<30%) and strong (>50%) yield reduction, respectively. This selection system can be applied in screening and breeding programmes to facilitate breeding for weed tolerance. No significant difference in weed tolerance was detected between six soybean genotypes of maturity groups 000 to 00. Morphological traits that might influence competitive ability, for example light absorption, leaf area and lateral shoots, were assessed, and their potential for indirect selection for weed tolerance is discussed.

to goods and services for residents. In 2013, a survey was conducted
to examine the retail planning practices of all the 110 small- and
medium-sized municipalities in Washington State. The online survey
addressed data collection and analysis, land use, retail recruitment,
inter-jurisdictional cooperation and evaluation. A large majority of
respondents had retail planning goals in their comprehensive plans,
yet only 30% dedicated staff for implementation. Jurisdictions would
benefit from regional retail planning (especially in light of changes
underway from e-commerce), yet the state tax structure places
municipalities in competition.

Soybean (Glycine max (L.) Merr.) is able to fix atmospheric nitrogen in symbiosis with the bacteria Bradyrhizobium japonicum. Because these bacteria are not native in European soils, soybean seeds must be inoculated with Bradyrhizobium strains before sowing to fix nitrogen and meet their yield potential. In Central Europe soybean cultivation is still quite new and breeding of early maturing soybean varieties adapted to cool growing conditions has just started.

Heightened interest in biologically-based methods of raising crop yields has driven research into beneficial microorganisms that can be used to improve crop growth and productivity. This review addresses the potential of rhizobia, vesicular-arbuscular mycorrhizae (VAM), and plant-growth-promoting rhizobacteria to improve growth of soybean (Glycine max (L.) Merr) under temperate conditions. Mechanisms of action of beneficial microorganisms and considerations for utilization at all root-zone temperatures (RZTs) are reviewed. Subsequent sections address current knowledge on the inhibition of soybean growth at low RZTs and the potential of beneficial microorganisms to alleviate low temperature stress. The three categories of beneficial microorganisms discussed differ in their modes of action and have shown potential for additive or synergistic growth promotion of soybean at all RZTs. At low RZT, pot and field studies have identified strains of rhizobia and PGPR, as well as certain phytohormones, that ameliorate the inhibitory effects of cold stress on soybean growth through a variety of mechanisms. Wider use of these treatments could aid the expansion of soybean cultivation in cold climates.

The effect of ploughing depth and mechanical soil loading on the performance of pea sole crops, oat sole crops and pea–oat intercrops was investigated in field experiments under organic farming conditions at two sites in Germany in 2009 and 2010. The two ploughing depths were short-term shallow ploughing to a soil depth of 7–10 cm and deep ploughing to 25–30 cm. Wheel loads of 26 and 45 kN, which correspond to typical rear wheel loads of field machinery used during sowing operations, were compared to an uncompacted control. Shallow ploughing resulted in a greater penetration resistance in the 14–28 cm soil layer compared to deep ploughing. An increase in mechanical soil loading intensity increased the bulk density and decreased the air capacity in the 10–15 cm soil layer, whereas the penetration resistance was not affected. The annual weed infestation in pea sole crops was higher after shallow than after deep ploughing at both sites. Pea–oat intercrops compensated for the higher weed infestation after shallow ploughing at one site due to their excellent weed suppressive ability. Dependent on oat productivity, pea–oat intercrops produced comparable or higher grain and protein yields than pea sole crops. Intercropped pea yield components and grain protein yields were significantly lower than those of sole cropped peas. The ploughing depth did not affect pea grain yields in either year and oat yields in 2009. Due to a better emergence, the grain and protein yield of sole and intercropped oats were significantly higher after shallow ploughing in 2010. Mechanical soil loading did not have any effect on the yield performance of pea sole crops, oat sole crops and pea–oat intercrops in 2009. In 2010, mechanical soil loading of 26 kN and 45 kN decreased the pea grain yield by 12.1% and 20.8% respectively, regardless of sole or intercropped. Neither the grain yield nor the grain quality of sole and intercropped oats was affected by the mechanical soil loading in 2010. Grain and crude protein yields of total crop stands decreased with increasing mechanical soil loading after deep ploughing, whereas no significant differences were revealed after shallow ploughing. The present study confirms the positive qualities of pea–oat intercrops with regard to weed suppression and plant performance. Shallow ploughing mitigates the risk of a decrease in crop performance caused by heavy field traffic and provides an alternative to deep ploughing even in low weed competitive organically farmed grain legumes.

The aim of the study was to establish a diagnostic tool to narrow down
the causes for pea yield depressions. A differential two-level
diagnostic test system was established under controlled conditions using
peas (Pisum sativum L.) as test plants. Soils from 22
organically managed sites with unexplained moderate to high pea yield
losses were tested in level 1 diagnostics (γ-irradiation to eliminate
potentially harmful organisms, nutrient additions to compensate for
potential nutrient deficiencies or activated charcoal amendment to bind
and thereby to immobilize potentially phytotoxic compounds). Results
showed that organisms harmful to the test plant were the primary cause
of limited germination and growth in most of the sampled soils, whereas a
positive effect of nutrient addition was rarely found and toxins were
not involved. Level 2 diagnostics (pesticides targeting ascomycetes,
oomycetes, Rhizoctonia spp., nematodes) further narrowed down the
organisms involved in yield depressions. Oomycetes were identified as
the primary reason for limited germination rates, and, in some soils,
also for limited growth of established seedlings. In other soils, a
multitude rather than a single group of pathogens was involved in
limited growth. Plant-pathogenic nematodes were never found to be
limiting for crop growth parameters. Harmful effects of pesticides were
found in several soils, hinting at an important role of beneficial soil
organisms in the suppression of pathogens causing yield depression in
legumes. The bioassay used in the present study was robust and could
thus serve as a low-cost tool for agricultural advisors and farmers to
predict the risk of yield losses in legumes and to narrow down causes,
helping them to develop appropriate strategies.

(EMG); der EMG-Sensor misst mittels auf der Haut aufgebrachten
Elektroden alle Potenzialänderungen, die bei der Kontraktion des
Kaumuskels (M. masseter) entstehen. Analyse und Bewertung der
aufgezeichneten EMG-Daten und Signalmuster finden mit einer
Analysesoftware statt, die die Dauer des Wiederkauens und der weiteren
Kauaktivitäten ausgibt. Dies lässt eine qualitative und quantitative
Bewertung auf Einzeltierbasis zu, die wiederum die Basis für eine
frühzeitige Krankheitserkennung bildet.

In eukaryotes, the poly(A) tail
added at the 3′ end of a mRNA precursor is essential for the regulation
of mRNA stability and the initiation of translation. Poly(A) polymerase
(PAP) is the enzyme that catalyzes the poly(A) addition reaction.
Multiple isoforms of PAP have been identified in vertebrates, which
originate from gene duplication, alternative splicing, or
post-translational modifications. The complexity of PAP isoforms
suggests that they might play different roles in the cell. Phylogenetic
studies indicate that vertebrate PAPs are grouped into three clades
termed α, β and γ, which originated from two gene duplication events. To
date, all the available PAP structures are from the PAPα clade. Here,
we present the crystal structure of the first representative of the PAPγ
clade, human PAPγ, bound to cordycepin triphosphate (3′dATP) and Ca2+.
The structure revealed that PAPγ closely resembles its PAPα ortholog.
An analysis of residue conservation reveals a conserved catalytic
binding pocket, whereas residues at the surface of the polymerase are
more divergent.

The objective of the study was to evaluate whether magnetic resonance
imaging (MRI) offers the opportunity to reliably analyze body
composition of pigs in vivo. Therefore, the relation between
areas of loin eye muscle and its back fat based on MRI images were used
to predict body composition values measured by dual energy X-ray
absorptiometry (DXA). During the study, a total of 77 pigs were studied
by MRI and DXA, with a BW ranging between 42 and 102 kg. The pigs
originated from different extensive or conventional breeds or crossbreds
such as Cerdo Iberico, Duroc, German Landrace, German Large White,
Hampshire and Pietrain. A Siemens Magnetom Open was used for MRI in the
thorax region between 13th and 14th vertebrae in order to measure the
loin eye area (MRI-LA) and the above back fat area (MRI-FA) of both body
sides, whereas a whole body scan was performed by DXA with a GE Lunar
DPX-IQ in order to measure the amount and percentage of fat tissue
(DXA-FM; DXA-%FM) and lean tissue mass (DXA-LM; DXA-%LM). A linear
single regression analysis was performed to quantify the linear
relationships between MRI- and DXA-derived traits. In addition, a
stepwise regression procedure was carried out to calculate (multiple)
regression equations between MRI and DXA variables (including BW).
Single regression analyses showed high relationships between DXA-%FM and
MRI-FA (R2 = 0.89, √MSE = 2.39%), DXA-FM and MRI-FA (R2 = 0.82, √MSE = 2757 g) and DXA-LM and MRI-LA (R2 = 0.82, √MSE = 4018 g). Only DXA-%LM and MRI-LA did not show any relationship (R2 = 0). As a result of the multiple regression analysis, DXA-LM and DXA-FM were both highly related to MRI-LA, MRI-FA and BW (R2 = 0.96; √MSE = 1784 g, and R2
= 0.95, √MSE = 1496 g). Therefore, it can be concluded that the use of
MRI-derived images provides exact information about important
‘carcass-traits’ in pigs and may be used to reliably predict the body
composition in vivo.

Rubber flooring in dairy cows has been discussed controversially,
because different studies found as well advantages as disadvantages
regarding claw health. Until today little is known regarding the effect
of rubber flooring on the reproductive performance of dairy cows.
Therefore, the study aimed at comparing fertility and the milk yield of
dairy cows on concrete or rubber flooring. Days to first breeding (DFB),
days open (DO), services per conception (SC), heat detection rate
(HDR), conception rate (first service; CR1st service) and the
corresponding 100 days in milk (DIM) milk yield were evaluated for 139
lactation periods of 91 cows of the University Munich Livestock Center
dairy herd. The cows entered the study as heifers and were housed on the
same flooring system during their whole productive life. All cows
underwent the same management, feeding, and climatic conditions. A mixed
model procedure was used to analyse data. Cows on rubber (RSF) compared
with cows on concrete flooring (CSF) showed less DFB (76.8/94.6; P<0.01), less DO (98.8/138.4; P<0.01), and no difference regarding SC (1.5/1.7; P=0.267).
Furthermore, no differences regarding the 100 DIM milk yield could be
established. The calculation of HDR (54.9%/27.9%) and CR1st service
(39.4/29.4) showed also high advantages for the rubber flooring. It was
concluded that rubber flooring affects an earlier onset of regular
oestrus behaviour and improves the economically sensible fertility trait
of calving interval by earlier pregnancy and less days open but does
not cause higher milk yield.

The improvement of carcass quality is one of the main breeding goals in
pig production. To select appropriate breeding animals, it is of major
concern to exactly and reliably analyze the body composition in vivo.
Therefore, the objective of the study was to examine whether the
combination of dual-energy X-ray absorptiometry (DXA) and magnetic
resonance imaging (MRI) offers the opportunity to reliably analyze
quantitative and qualitative body composition characteristics of
different pig breeding groups in vivo. In
this study, a total of 77 pigs were studied by DXA and MRI at an average
age of 154 days. The pigs originated from different autochthonous or
conventional breeds or crossbreeds and were grouped into six breed
types: Cerdo Ibérico (Ib); Duroc × Ib (Du_Ib); White Sow Lines (WSL,
including German Landrace and German Large White); Hampshire/Pietrain
(Pi_Ha, including Hampshire, Pietrain × Hampshire (PiHa) and Pietrain ×
PiHa); Pietrain/Duroc (Pi_Du, including Pietrain × Duroc (PiDu) and
Pietrain × PiDu); crossbred WSL (PiDu_WSL, including Pietrain × WSL and
PiDu × WSL). A whole-body scan was performed by DXA with a GE Lunar
DPX-IQ in order to measure the amount and percentage of fat tissue (FM;
%FM), lean tissue (LM; %LM) and bone mineral, whereas a Siemens Magnetom
Open with a large body coil was used for MRI in the thorax region
between 13th and 14th vertebrae in order to measure the area of the loin
(LA) and the above back fat area (FA) of both body sides. A GLM
procedure using SAS 9.2 was used to analyze the data. As expected, the
native breed Ib followed by Du_Ib crossbreeds showed the highest %FM
(27.2%, 25.0%) combined with the smallest LA (46.2 cm2, 73.6 cm2),
whereas Ib had the lowest BW at an average age of 154 days. Pigs with
Pi_Ha origin presented the least %FM (12.4%) and largest LA (99.5 cm2).
The WSL and PiDu_WSL showed an intermediate body composition.
Therefore, it could be concluded that DXA and MRI and especially their
combination are very suitable methods to reliably identify differences
in body composition and carcass traits among different pig lines in vivo.

A sensitivity analysis for a well-established baseflow separation
technique, a two parameter recursive digital filter, is presented. The
sensitivity of the calculated baseflow index to errors or uncertainties
of the two filter parameters and of the initial baseflow value is
analytically ascertained. It is found that the influence of the initial
baseflow value is negligible for long time series. The propagation of
errors or uncertainties of the two filter parameters into the baseflow
index is expressed by a dimensionless sensitivity index, the ratio
between the relative error of the baseflow index and the relative error
of the respective parameter. Representative index values are derived by
application of the resulting equations to 65 North American catchments.
In the mean the parameter a, the recession constant, has a stronger influence on the calculated baseflow index than the second filter parameter BFImax. This is favourable in that a
can be determined by a recession analysis and therefore should be less
uncertain. Whether this finding also applies for a specific catchment
can easily be checked by means of the derived equations.

Baseflow indices for 65 North American catchments are compared, which
were calculated with seven different baseflow separation methods
(HYSEP1, HYSEP2, HYSEP3, PART, BFLOW, UKIH, and Eckhardt). Since the
true values of the baseflow index are unknown, it cannot be said, which
one of the methods gives the best estimates. However, the results of the
Eckhardt method appear to be hydrologically more plausible than those
of the other algorithms. Furthermore, this algorithm can be applied to hydrographs
of any time step length. A simple technique for assessing the recession
constant – one of the parameters of the algorithm – is presented.

Here, we report the design of unprecedented, non-FRET based
cGMP-biosensors, named FlincGs, to assess the dynamics of nitric oxide
(NO) and atrial natriuretic peptide (ANP) induced synthesis of
intracellular cGMP, [cGMP](i). Regulatory fragments of PKG I alpha, PKG I
beta, and an N-terminal deletion mutant of PKG I alpha were fused to
circular permutated EGFP to generate alpha-, beta-, and delta-FlincG,
with high dynamic ranges and apparent K(D,cGMP) values of 35 nM, 1.1
microM, and 170 nM, respectively. All indicators displayed significant
selectivity for cGMP over cAMP, and 1.5- to 2.1-fold increases in
fluorescence intensity at 510 nm when excited at 480 nm. Surprisingly,
FlincGs displayed an additional excitation peak at 410 nm. delta-FlincG
permitted ratiometric (480/410 nm) measurements, with a cGMP-specific
3.5-fold ratio change. In addition, delta-FlincG presented cGMP
association and dissociation kinetics sufficiently fast to monitor rapid
changes of [cGMP](i) in intact cells. In unpassaged, adenoviral
transfected vascular smooth muscle (VSM) cells, delta-FlincG had an
EC(50,cGMP) of 150 nM, and revealed transient global cGMP elevations to
sustained physiological NO (EC(50,DEA/NO) = 4 nM), and the decay phase

Ziel der vorliegenden Studie war es, den Einfluss eines elastischen Bodens im Laufstall auf Milchleistung, Fettgehalt, Fettmenge, Eiweißgehalt, Eiweißmenge und Zellzahlen zu untersuchen. Hierzu wurden 49 Kühe in 53 Laktationen (jew. 305 d) in zwei Gruppen aufgeteilt. Die Versuchsgruppe (EBG) (n = 26) wurde auf Betonspaltenboden mit elastischen Auflagen, die Kontrollgruppe (BBG) (n = 27) auf Betonspaltenboden ohne Auflagen aufgestallt. Beide Abteile unterschieden sich durch die Art der Liegeboxen. Der EBG standen Hochboxen, der BBG Tiefboxen zur Verfügung. Zur Auswertung kamen Daten der Milchleistungsprüfung des Landeskuratorium der Erzeugerringe für tierische Veredelung in Bayern e.V. und Daten von Bewegungszählern, die die Tiere am Halsband trugen. Die Auswertung ergab ein signifikant höheres Aktivitätslevel der EBG gegenüber der BBG. Bei der Auswertung der Milchdaten lagen keine signifikanten Unterschiede bei Milchmenge, Eiweißgehalt und Eiweißmenge vor. Fettgehalt und Fettmenge lagen in der BBG signifikant höher als in der EBG. Der Zellgehalt lag in der EBG signifikant niedriger als in der BBG. Eine Auswertung getrennt nach Aktivitätslevel zeigte einen Trend abnehmender Leistung mit steigender Aktivität. Demnach konnte im Rahmen dieser Studie keine Leistungsverbesserung der Tiere durch den Einfluss des elastischen Bodens bewiesen werden. Das signifikant höhere Aktivitätslevel der EBG spricht aber für ein erhöhtes Maß an Kuhkomfort durch den weichen Boden.

Models used for prognostic purposes should, as far as possible, be physically based, as empirical modelslose their validity if boundary conditions are changed too much. Yet, a physical description of hydrological processes may be possible at the scale of single pores, but not in models for entire catchments. In the lattercase, in general small-scale physics developed for homogeneous systems is used under the assumption that the corresponding equations can be applied at larger spatial scales with the same parameters. An example is Darcy’s law describing theflow in porous media. Originally, it was developed from experiments with small soilcores. In numerical groundwater models, however, it is applied at scales of up to kilometres. At these scales,parameters like the hydraulic conductivity are no longer measurable and therefore are considerably uncertain.This pertains to a wide range of models, not only to numerical groundwater models such as MODFLOW(McDonald and Harbaugh, 1989), but also to analytical models that simulate processes at the catchment scalesuch as SHE (Abbottet al., 1986) or SWAT (Arnoldet al., 1998).

There is widespread agreement that good correspondence of measured and calculated streamflow at the catchment outlet is not a sufficient criterion for the validity of a physically based hydrologic model, but that additional knowledge concerning catchment-internal processes is needed (e.g. Beven, 1989). Yet, for many catchments such information is not available and the model’s performance can only be assessed by comparing calculated and measured streamflow. However, the information provided by a hydrograph is not limited to the absolute height of the runoff. For example, a separation of the hydrograph allows for identification of runoff components originating from different compartments of the considered system, and responding differently delayed and smoothed to rainfall events. General Circulation Models
simulate significant changes of temperature and precipitation over
Europe as part of the anthropogenic climate change. In this study, the
impacts of climate change on groundwater recharge and streamflow
in a central European low mountain range catchment are investigated
using a conceptual eco-hydrologic model, a revised version of the Soil
and Water Assessment Tool (SWAT). To improve the reliability of our
simulations, we compile plant physiological studies concerning the
influence of elevated ambient CO2 concentrations on stomatal conductance
and leaf area. Using this information to parameterise the model, we
evaluate the impacts of two climate change scenarios, which represent a
wide range of assumptions concerning future greenhouse gas
emissions and climate sensitivity. The resulting effects on mean annual
groundwater recharge and streamflow are small, as increased atmospheric
CO2 levels reduce stomatal conductance thus counteracting increasing potential evapotranspiration
induced by the temperature rise and decreasing precipitation. There
are, however, more pronounced changes associated with the mean annual
cycle of groundwater recharge and streamflow. Our results imply that due
to the warming a smaller proportion of the winter precipitation will
fall as snow. The spring snowmelt
peak therefore is reduced while the flood risk in winter will probably
increase. In summer, mean monthly groundwater recharge and streamflow
are reduced by up to 50% potentially leading to problems concerning
water quality, groundwater withdrawals and hydropower generation.

Uncertainty in parameters characterising different land covers leads to uncertainty in model predictions of land use change
effects. In this study, a new approach is presented which allows a
model to be assessed to see whether it is suitable for investigating
land use change scenarios in the sense that different land covers can be
significantly distinguished in their effects on model output. It
consists of the following steps: (a) The uncertainty in land
cover-dependent parameters is quantified. (b) The model of an artificial
catchment with representative characteristics and uniform land cover is
established. (c) Using this artificial catchment, Monte Carlo
simulations are carried out to determine the uncertainty in the model
response to different land covers. (d) By comparing the results for two
covers, respectively, a dimensionless test statistic, the distinction
level, is calculated. The distinction level is a normalised probability
that two independent realisations of land covers which are parameterised
within their range of natural uncertainty will yield distinct model
responses. If the distinction level is greater than or equal to 90%, the
land covers are assumed to have a significantly different effect on the
model output. An example of the application of the new method is
provided using the eco-hydrologic model SWAT-G. The land covers forest,
pasture and arable land can be significantly distinguished by their long-term means of surface runoff, groundwater recharge and streamflow. The minimum proportion of the catchment area
on which land cover must change in order to obtain significantly
distinct model responses depends on the land covers involved and the
considered hydrologic variable. In the case of a change between pasture
and forest and with regard to average streamflow, this minimum
proportion amounts to about 25%, a value that compares well with the
results of paired catchment studies.

Biological sulfide oxidation is a reaction occurring in all three
domains of life. One enzyme responsible for this reaction in many
bacteria has been identified as sulfide:quinone oxidoreductase (SQR).
The enzyme from Rhodobacter capsulatus is a peripherally membrane-bound
flavoprotein with a molecular mass of approximately 48 kDa, presumably
acting as a homodimer. In this work, SQR from Rb. capsulatus has been
modified with an N-terminal His tag and heterologously expressed in and
purified from Escherichia coli. Three cysteine residues have been shown
to be essential for the reductive half-reaction by site-directed
mutagenesis. The catalytic activity has been nearly completely abolished
after mutation of each of the cysteines to serine. A decrease in
fluorescence on reduction by sulfide as observed for the wild-type
enzyme has not been observed for any of the mutated enzymes. Mutation of
a conserved valine residue to aspartate within the third flavin-binding
domain led to a drastically reduced substrate affinity, for both
sulfide and quinone. Two conserved histidine residues have been mutated
individually to alanine. Both of the resulting enzymes exhibited a shift
in the pH dependence of the SQR reaction. Polysulfide has been
identified as a primary reaction product using spectroscopic and
chromatographic methods. On the basis of these data, reaction mechanisms
for sulfide-dependent reduction and quinone-dependent oxidation of the
enzyme and for the formation of polysulfide are proposed.

Die Kalibrierung eines Modells ermöglicht eine Reduzierung der Parameterunsicherheit und damit der Unsicherheit in Simulationsergebnissen. Komplexe Modelle können mittels stochastischer globaler Optimierungsalgorithmen automatisch kalibriert werden. Zwei dieser Algorithmen werden hinsichtlich ihrer Leistungsfähigkeit miteinander verglichen: einerseits der Boender-Timmer-Rinooy Kan (BTRK) Algorithmus, ein Multistart-Algorithmus mit vorhergehender Clusterung, und andererseits der Shuffled Complex Evolution (SCE) Algorithmus, ein Genetischer Algorithmus. Die beiden Algorithmen werden auf ein analytisches verteiltes Modell zur Nachbildung des Wasser- und Stoffhaushalts in meso- bis makroskaligen Einzugsgebieten oberirdischer Gewässer angewendet, die Version SWAT-G des Soil and Water Assessment Tool (SWAT). Der SCE Algorithmus erweist sich dem BTRK Algorithmus als überlegen. Es wird eine hohe Güte der Modellanpassung erreicht.

The Soil and Water Assessment Tool (SWAT) is a well established
distributed eco-hydrologic model. However, using the example of a
mesoscale catchment in Germany it is shown that the version SWAT99.2 is
not able to correctly reproduce the runoff generation in a low mountain
region. The calculated contribution of the baseflow to the streamflow is
far too high whereas the interflow is strongly underestimated.
Alternatively, the modified version SWAT-G can be used which, as is
demonstrated in this paper, yields far better results for catchments
with predominantly steep slopes and shallow soils over hard rock
aquifers. In the example, calibrating the model over three hydrologic
years of daily streamflow, the model efficiency increases from −0.17 to
+0.76. The modifications in SWAT-G allow hydrological processes to be
modelled in low mountain ranges while not restraining the applicability
of the model to catchments with other characteristics.

Parameters of hydrologic models
often are not exactly known and therefore have to be determined by
calibration. A manual calibration depends on the subjective assessment
of the modeler and can be very time-consuming though. Methods of
automatic calibration can improve these shortcomings. Yet, the high
number of parameters in distributed models makes special demands on the
optimization. In this paper a strategy of imposing constraints on the
parameters to limit the number of independently calibrated values is
outlined. Subsequently, an automatic calibration of the version SWAT-G
of the model SWAT (Soil and Water Assessment Tool) with a stochastic
global optimization algorithm, the Shuffled Complex Evolution algorithm,
is presented for a mesoscale catchment.

In fattening bulls tail tip alterations are described as health issues
associated with (sub)acute rumen acidosis and lameness. Investigations
of dairy cows’ tail tips are scarce; however, there is evidence that
tail tip alterations occur as a result of intensive feeding management
and metabolic imbalance. In June 2020, we investigated 68 German
Holstein cows for tail tip alterations using a handheld thermal camera
(FLIR® T1030). Thermal images of the shaved tail tip were taken from two
different perspectives: (p1) tail in front of the udder and (p2) held
tail in front of the ground. Additionally, all cows were evaluated for
any kind of tail tip alterations, body condition score (BCS), and
locomotion score (LMS). Milk yield data, resulting from latest
performance testing were collected, too. Images were analysed using FLIR
Tool+ and a generalised linear model was used to correct measured
temperatures for fixed effects. The prevalence of tail tip alterations,
regardless the findings, was 94%. Raw temperature data showed high
correlations of r² = 0.79 for maximum and average temperature and r² =
0.70 for minimum temperature between p1 and p2 (P < 0.001).
Tail tip necroses and BCS > 3 decreased the tail’s temperature, while
swelling of the tail tip increased its temperature. Average, maximum
and minimum temperatures for perspective 1 and 2 were 34.05 (±0.16) and
33.78 (±0.20) °C, 35.46 (±0.11) and 35.52 (±0.11) °C, and 30.30 (±0.30)
and 29.22 (±0.38) °C, respectively. Temperatures of necrotic tissues (n =
4) decreased on average by 8.38 °C compared to the average temperature
and were close to the minimum. We concluded thermal imaging can be used
for the detection of alterations regarding the tail’s temperature, which
could allow an early-warning system for swellings, probably caused by
inflammation and possibly resulting in necroses. Further studies are
needed to evaluate the praevalence of tail tip alterations in dairy cows
on larger samples and to clarify their etiology. However, thermal
imaging can be used as an early-warning system for tail alterations.
Each early-warning system regarding health issues, improves animal
welfare as it allows adapting management strategies early.

Research
investigating necrotic tail tips in dairy cows resulting in necrotic tissues is
scarce. However, there is evidence that in dairy cattle tail tip necroses exist
with high prevalence. In piglets, the latest research described tail and ear
necroses not because of tail and ear biting only, but because of swine inflammation
and necrosis syndrome (SINS). Besides tails and ears, SINS includes
inflammation of claws, heels, and teats. In cattle, tail tip necroses are described
mainly in fattening bulls. As known so far, these findings are often discussed
related to slatted flooring, intensive housing systems and management
strategies. However, an association with subacute rumen acidosis and laminitis is
described. In order to
investigate what kind of and how often tail tip alterations appear in dairy
cattle, data of 87 German Holstein dairy cows were collected over a period of
12 months. All cows were evaluated for tail tip alterations, body condition
score (BCS), and locomotion score (LMS) every two weeks. In addition, milk
yield data resulting from performance testing were included. Thermographic images
of the tails were taken once. Firstly, all kind of tail tip alterations were
described and collected. After 6 months, we categorized the observed
alterations and developed a tail scoring system. The scoring for each specified
trait (tail tip, tail ring) ranged from 0 to 4. The overall
prevalence for tail alterations was 94%. Especially tail tip alterations had a constantly
high prevalence of 56%. Cows affected by an increased average tail tip score,
showed higher locomotion scores compared to others (P = 0.02). The prevalence of ring-like tail alterations increased
from first to second lactation cows from 9 to 46%. Regarding the BCS, lighter
cows showed higher scores due to ring-like alterations than heavier cows (P = 0.054). The most often occurring
anomalies of the tail were sports or scurf (21.6%), followed by verruca-like
mass (10.2%), swelling (8.4%), and thinning (4.3%). The results
and especially the scoring system can serve as a template for further studies considering
larger samples sizes, to investigate prevalence for tail necroses and other
tail anomalies in different herds and management systems. It was hypothesized,
that an inflammatory condition in dairy cows showing up in altered/necrotic
tail tips or rings exists, which is in relationship with claw disorders indicated
by lameness. If so, the tail score of a cow could be used as health indicator
to evaluate the health status in dairy production systems.
